Presentation title: Expression and Purification of the Antimicrobial Peptide, Subtilin, from E.coli.
Abstract: Subtilin is a member of the lantibiotic antimicrobial peptide family and is naturally produced by B. subtilis bacteria. It can be used to inhibit bacterial growth and can assist in wound healing. We chose to expression subtilin in E. coli in order to purify it and use it as a topical wound treatment. We created a DNA plasmid with an inducible promoter, transformed it into E. coli and turned on the gene circuit using IPTG. Next, we purified subtilin from the other bacterial proteins using affinity chromatography and a Ni2+-column. Our final step will be to test the antimicrobial effectiveness of the purified peptide on Gram-positive bacteria.
